Brian O'Conner (Paul Walker) viene scarcerato perché torni nuovamente sotto copertura e s'infiltri nella banda di un pericoloso trafficante di droga di Los Angeles noto come Braga. Ma in questa sua nuova missione Brian dovrà fare coppia proprio con colui che aveva ingannato anni prima: Dominic Toretto (Vin Diesel). Brian e Dominic dovranno imparare nuovamente a fidarsi l'uno dell'altro e dovranno cercare d'incastrare Braga con l'aiuto di un informatore, James Park.
